Therese Ann Rutherford was born on November 2, 1920 in Vancouver, British Columbia, Canada to a former Metropolitan Opera tenor and actress. At an early age, she performed in various radio shows, before making her film debut in "Waterfront Lady" in 1935. Between 1935 and 1937, she appeared in eighteen films and was a popular leading lady in Westerns with Gene Autry and John Wayne. In 1937, she landed her first big role as Andy Hardy's girlfriend, Polly Benedict, in You're Only Young Once. The popular film series ran until 1942 with Ann playing Polly twelve times, her last appearance was in Andy Hardy's Double Life. Ann was placed under contract with MGM where she appeared in such films as A Christmas Carol (1938) and Pride and Prejudice (1940). In 1939, she was loaned to Selznick International to appear as Carreen O'Hara in Gone with the Wind (1939).

Ann also starred in a series of mystery/comedies with Red Skelton; Whistling in the Dark (1941), Whistling in Dixie (1942), and Whistling in Brooklyn (1943). After making Washington Melodrama (1941), Ann left MGM to freelance in the early 1940s, starring in such films as Orchestra Wives (1942), Two O'Clock Courage (1945) and The Secret Life of Walter Mitty (1947). She retired from films in 1950, but returned to her old studio in 1972 to make They Only Kill Their Masters, which was shot on the old Andy Hardy set. In the 1970s she also made two guest appearances on The Bob Newhart Show, where she played Aggie Harrison, Emily Hartley's mother. Ann could have made a comeback in 1997 by playing Rose Calvert in Titanic, but she turned down the role, and it went to Gloria Stuart.

Ann continues thrill fans by attending events honoring Gone With the Wind. In October 2004, she attended the Margaret Mitchell Birthday celebration in Jonesboro, Georgia,where she signed autographs and reminisced about making the movie. In 2007, she was the guest star at the Marietta, Georgia Gone With the Wind Museum's "The Heart and History of Hollywood" event emceed by TCM host Robert Osborne. As of December 2011, Ann's remaining co-stars from Gone With the Wind include; Olivia DeHavilland, Alicia Rhett, Mickey Kuhn, Patrick Curtis, Mary Anderson and Rick Holt.

Rutherford was married twice; on Christmas Eve, 1942, she married David May. The couple had a daughter before divorcing in 1953. Ann then married William Dozier (who produced the Batman TV series) and they had one child before his death in 1991. Ann has a star on the Hollywood Walk of Fame and in 1988, won the Golden Boot Award which honors actors and actresses who star in Westerns.
